Assassin's Creed: Brotherhood הינו המשכו של המשחק Assassin's Creed 2, מבחינה כרונולוגית ומבחינה עלילתית גם כן, כאשר גיבור המשחק חוזר להיות אציו המוכר לנו מהמשחקים הקודמים.
המשחק הינו הראשון בסדרה המאפשר לשחק עם שחקנים נוספים דרך רשת האינטרנט (מולטיפלייר).
